Spurs are believed to be paying an estimated £15 million for the return of the 26-year-old striker.
"We are delighted to announce that we have reached agreement with Portsmouth for the transfer of Jermain Defoe," a statement from Spurs has said.
"Personal terms have been agreed and the striker will today undergo a medical."
